Description

The clinical utility of cisplatin, carboplatin or oxaliplatin and other Pt(II)-based anticancer agents is hampered by rapid deactivation and severe side effects. This project proposes the synthesis of lipophilic Pt(IV) prodrugs—octahedral complexes that remain inactive until reduced in the hypoxic, glutathione-rich environment of cancer cells. By functionalizing the axial positions with long-chain fatty acids, we will promote stable encapsulation within the hydrophobic bilayer of engineered nanovesicles, leveraging "stealth" delivery to bypass traditional resistance mechanisms.
